Anfang der 70er macht sich ein Trupp von Soldaten, Regierungsbeauftragen und Zivilisten auf, eine mysteriöse Insel zu erkunden. Die Gruppe unter dem Kommando von Lieutenant Colonel Packard (Samuel L. Jackson), zu der auch der mysteriöse „Reiseleiter“ Bill Randa (John Goodman), der Spezialkräfte-Veteran James Conrad (Tom Hiddleston) und die Kriegsfotografin Mason Weaver (Brie Larson) gehören, wird sehr unfreundlich empfangen: Etwas holt die Helikopter vom Himmel! Die Überlebenden der Abstürze erfahren, dass auf „Skull Island“ ein Riesenaffe haust, eine gefährliche Kreatur, die wie ein einsamer Gott über die Insel streift. Doch viel Zeit zum Lernen bleibt nicht, denn als die Expeditionstruppe auf Eingeborene und den geheimnisvollen, schon vor langer Zeit auf der Insel gestrandeten Amerikaner Hank Marlow (John C. Reilly) trifft, erfährt sie, dass Affe Kong zwar der König der Insel ist, aber noch viele weitere Monster im Dickicht lauern…
